Abstract:

In conjunction with two large mesocosm experiments, sub-samples were analyzed to determine the impact of oil, dispersant and dispersed oil on the bacterial community. Using 16S rRNA gene tags, it was determined that the bacterial community responded differently to oil in the presence of dispersant or to dispersant alone. The response timing and extent varied between the two experiments indicating the initial conditions have a large impact on the response of the community. Purpose:

To document the response of the bacterial community present in coastal seawater when exposed to oil, dispersant and dispersed oil.

Data Parameters and Units:

16S rRNA sequence data submitted to NCBI at http://www.ncbi.nlm.nih.gov/Traces/sra/sra.cgi?study=SRP020514. Sequence data: 106 runs, run name, BioSample, Sample Name, MBases, MBytes, Experiment, Replicate, SRA Sample, Timepoint, Collection Date, Geographic Location, Treatment.

Methods:

Sequence data was obtained after filtering 50 ml of water through a 0.22 μm filter using a syringe. Filters were stored frozen. DNA was extracted following a standard phenol/chloroform procedure and the V6 region of the 16S rRNA gene was amplified using a pool of primers modified to include the Ion Torren adapter sequences. Sequencing was carried out on an Ion Torrent PGM. Sequence data was filtered for quality and analyzed using QIIME.
