Abstract:

This dataset will include fertility potential assays using ELISA’s from blood serum samples. Concentrations of hormones will be reported as pg/ml of serum.

Purpose:

Monitor levels of Inhibin A, Inhibin B, and Anti-Mullerian Hormone (ng/ml) to determine impacts on fertility potential.

Methods:

Sample Collection: Serum Collect 7.5 mL whole blood into a marbletop serum separator tube. Invert a few times and allow to clot on ice for 30 minutes. Spin at 2700 rpm for 15 minutes. Remove supernatant and place in a red top cryovial. Store in -20 freezer or in LN2. When samples are removed and thawed for the first time they are aliquotted out to prevent numerous freeze/thaw cycles Sample Processing: Samples are run according to manufacturer's protocol.

Instruments:

DS2 Automated ELISA instrument by Dynex Technologies Inhibin A and Inhibin B human ELISAs by Beckman-Coulter Activin B Mouse ELISA by Cusabio.

Error Analysis:

Standard curves and no substrate blanks are run with each plate. High and low controls are included on Inhibin A and Inhibin B plates Samples are run in duplicate Sample %CV's are calculated and outliers are removed such that %CV is less than 20%
