Abstract:

Biodegradation assays with dibenzo[a,e]pyrene (DB[a,e]P) were conducted in the presence of a consortium of microbes isolated from oil-contaminated sediment. Sediments were collected from the Gulf of Mexico (29.895448 N 88.827780 W) on 2018-05-07. Data includes high throughput sequencing data demonstrating the changes to the individual microbial isolates as a result of DB[a,e]P contamination.

Purpose:

The purpose of the study was to determine the effects of dibenzo[a,e]pyrene on individual microbial isolates in biodegradation culture assays.

Methods:

A consortium of individual microbes were isolated from Gulf of Mexico sediment contaminated by the Deepwater Horizon oil spill in order to determine if isolates were able to degrade DB[a,e]P and how they changed as a result of the high molecular weight polycyclic aromatic hydrocarbon (HMW-PAH) contamination. Culture experiments were conducted in a brackish artificial seawater media (M10b: 4.0 g/L tryptone, 2.5 g/L yeast extract in artificial seawater) amended with (or without) 1 ppm DB[a,e]P for 28 days. Total DNA of the culture was extracted at the conclusion of the experiment and sequenced by Illumina MiSeq technology.
