Abstract:

Biodegradation assays with benzo[a]pyrene (BaP) were conducted in the presence of a consortium of microbes isolated from oil-contaminated sediment. Sediments were collected from the Gulf of Mexico (29.895448 N 88.827780 W) on 2018-05-07. Data includes high throughput sequencing data demonstrating the changes to the individual microbial isolates as a result of BaP contamination.

Purpose:

The purpose of the study was to determine the effects of benzo[a]pyrene on individual microbial isolates in biodegradation culture assays.

Methods:

A consortium of individual microbes were isolated from Gulf of Mexico sediment contaminated by the Deepwater Horizon oil spill in order to determine if isolates were able to degrade BaP and how they changed as a result of the high molecular weight-polycyclic aromatic hydrocarbon (HMW-PAH) contamination. Culture experiments were conducted in a brackish artificial seawater media (M10b: 4.0 g/L tryptone, 2.5 g/L yeast extract in artificial seawater) amended with (or without) 1 ppm BaP for 28 days. Total DNA of the culture was extracted at the conclusion of the experiment and sequenced by Illumina MiSeq technology.
