Abstract:

The dataset provides information about total circulating antioxidant capacity of seaside sparrows (Ammospiza maritima) collected from various sites in southeastern Louisiana from 2013 to 2015.

Purpose:

To test whether birds exposed to Deepwater Horizon oil differed from control birds in the ability of their plasma to counter an oxidative threat, as it could arise from exposure to polycyclic aromatic hydrocarbons (PAHs) from the oil spill.

Methods:

To determine the concentration of reactive oxygen metabolites (OXY), we used the OXY-Adsorbent test kit (Diacron International, Grosseto, Italy), following the manufacturer’s instructions, with minor modifications according to Costantini et al. (2011). The assay quantifies the ability of non-enzymatic antioxidants in a sample to face the pro-oxidant action of hypochlorous acid (HOCl). Samples were diluted 1:100 and combined with a reagent from the kit, then incubated (10 min at 37 °C). After incubation, a chromogen reagent was added to the solution and the absorbance was read in a spectrophotometer at 490 nm. Values were calibrated using a reference standard and converted to µmol/mL of HOCl neutralized. To determine the concentration of reactive oxygen metabolites (ROMs), we used the d-ROMs Test, a commercial kit (Diacron International, Grosseto, Italy), following the manufacturer’s instructions, with minor modifications according to Costantini et al. (2011). Samples were combined with reagents from the kit, then incubated (75 min at 37 °C). At the end of the incubation, a chromogen was added and absorbance values were read in a spectrophotometer at 505 nm. Values were calibrated using a reference standard and converted to millimolar of H2O2 equivalents.
