Abstract:

Mesocosm experiments were performed with seawater collected from Galveston Bay, Texas. Targeted polycyclic aromatic hydrocarbons (PAH) metabolites were measured in filtered samples from two treatments: 1) control (seawater) and 2) Diluted Chemically Enhanced Water-Accommodated Fraction (DCEWAF) of crude oil. The extracted samples were analyzed on high performance liquid chromatography triple quadrupole mass spectrometry (LC-MS/MS).

Purpose:

To determine the concentration of metabolites form during microbial oxidation of crude oil components.

Methods:

Dissolved phase was collected for each sample passing through a filtration series (GF/D and then 0.2µm). The dissolved phase for each sample was then acidified to pH 2 and extracted with Agilent’s PPL solid-phase extraction cartridges. The extracted samples were analyzed on high performance liquid chromatography triple quadrupole mass spectrometry (LC-MS/MS). Triplicate samples were collected for each treatment at 4 time intervals: Day 0, 4, 8, and 16.
