Abstract:

A mesocosm scale experiment was conducted to monitor the temporal changes in microbial diversity of microaggregates that form in response to the presence of oil and chemically dispersed oil. Fluorescent labeled probes were developed to specifically target microorganisms of interest to better understand the microbial structure and evolution of microaggregates.

Purpose:

To quantify the abundance of specific taxa within oil-microbe aggregates which form in seawater after exposure to oil and chemically dispersed oil.

Data Parameters and Units:

The headers are Treatment, Timepoint, Number of single cells (Alteromonas), Number of tagged single cells (Alteromonas), Number of aggregated cells (Alteromonas), Number of aggregated tagged cells (Alteromonas), Number of single cells (Alcanivorax), Number of tagged single cells (Alcanivorax), Number of aggregated cells (Alcanivorax), and Number of aggregated tagged cells (Alcanivorax).

Methods:

Water accommodated fraction (WAF) was prepared by adding 25mL of Macondo surrogate oil into 130L of seawater and allowing it to mix for 4 hours. For DCEWAF, Corexit was mixed with oil in a ratio of 1:20 and then 25mL of this mixture was added to 130L of seawater and mixed for 4 hours. The oil concentration for this experiment was determined as estimate oil equivalents and found to be 2.15 mg/L for WAF and 2.62 mg/L for DCEWAF. The oil used was Macondo Surrogate Crude oil.
