Abstract:

Sea surface microlayer and subsurface water samples were collected on polycarbonate membrane filters at the Looe Key coral reef and near Lois Key in a seagrass area in the Florida Keys. Samples were collected on July 29th through August 2nd, 2019. This dataset contains calculations of the abundance of all bacteria on the 220 samples from next-generation sequencing data. Results of bacteria abundance were obtained via sequencing on the Illumina MiSeq at Argonne National Laboratory and analysis was performed using QIIME2. Confidence intervals were calculated using the Wald calculation based off of a negative binomial distribution.

Purpose:

Analyze surfactant-associated bacteria abundance in the sea surface microlayer above a coral reef area and in a coastal seagrass area, to compare with results from previous experiments conducted in the Gulf of Mexico in oil slicks.

Data Parameters and Units:

The file ASV_File.xlsx contains the number of ASV hits of all bacterial genera and unassigned found in all samples. It includes - Accession Number (the number associated with run submitted to NCBI, only one run with all samples, so one accession number); SampleID = sample number; Latitude, longitude = exact location of sample collection in decimal degrees; Depth = depth in the water column that sample was collected from (surface: sea surface microlayer; 0.2m: 0.2 m below the surface; air: sample held in air for control); Date collected = date that sample was collected on (M/DD/YY); Column E, rows 1-6 = taxonomic ranks that correspond to bacteria taxonomy in following columns (Kingdom, Phylum, Class, Order, Family, and Genus). The file CIs.xlsx shows the calculation of the average number of ASV hits and confidence interval using the negative binomial Wald formula for each genus of interest. It includes - Species name; Site numbers (1-7); SML (surface) = sea surface microlayer used in calculation, average count of ASV hits for each site for this type, surface: depth of sample at surface of the ocean; SSW = subsurface water samples used in calculation, average count of ASV hits for each site for this type; 0.2m: depth of sample at 0.2 m below surface of the ocean; AC = Air control samples used in calculation, average count of ASV hits for each site for this type, air: depth of sample held in air; T = Student’s distribution coefficient for 95%; T/2 = Student’s distribution coefficient divided by two for use in negative binomial formula; N = number of samples; MIN_CI = minimum confidence interval calculated using the negative binomial formula; MAX_CI = maximum confidence interval calculated using the negative binomial formula. Note: NE (non-exposed) and NTC (non-template control) = controls. The dataset also includes a readme file.

Methods:

The sea surface microlayer sampling is conducted with a polycarbonate filter, placed on the sea surface away from the boat hull and from its hydrodynamic wake. The analysis is conducted with the DNA next-generation sequencing methods using the Illumina MiSeq to identify surfactant/oil associated bacteria.
