Abstract:
This project was undertaken to evaluate the viability, fertility, hatch success and larval survival of Florida Pompano fish upon exposure to chemically enhanced water accommodated fraction (CEWAF) of south Louisiana crude oil and Corexit 9500. At least one parent of the adult fish were similarly exposed to CEWAF of south Louisiana crude oil and Corexit 9500. Hence, this generation of fish were exposed to CEWAF in tanks for a specified period of time and evaluated in terms of total viable eggs, percentage of eggs that were fertilized, the percentage of fertilized eggs that hatched and the percentage of survival.
Data Parameters and Units:
Contains multiple worksheets that detail the experimental setup and conditions (Test Conditions table, Tank Descriptions), how stock and diluted CEWAF were made (Stock CEWAF Preparation, CEWAF Dilution Preparation respectively), and the mean total eggs, mean total viable eggs, mean fertilization (%), mean hatch (%), mean 3-day larval survival (%) after exposure to CEWAF (Hatch and Survival).
The worksheet “Test Conditions table”: Contains information regarding the setup of the experiments in terms of species, life stage of species, exposure type, renewal frequency, number of replicates per treatment, description of treatments, flow rates, aeration, and family cross details.
The worksheet “Tank Descriptions”: Tank (name of tank), Treatment (type of experiment- either a control or actual experiment), Start Date and End Date (beginning and conclusion of experiment; MM,DD,YYYY), Time Spike A and Date Spike A (time and date of initial addition of chemicals; MM,DD,YYYY; HH:MM), Time Clean SW A (time of chemical addition completion HH:MM), Time Spike B and Date Spike B (time and date of second addition of chemicals; MM,DD,YYYY; HH:MM), Time Clean SW B (time of chemical addition completion HH:MM), Notes (which cross of fish were in each tank).
The worksheet “Stock CEWAF Preparation”: Date and Start time (referring to when CEWAF was made; DD/MM/YYYY; HH:MM), Carboy ID, Oil (the amount of oil added to mixture, g), Corexit (the amount of Corexit added to mixture, g) and Seawater (quantity of seawater L), End Date and End Stir Time (referring to when CEWAF was made; DD/MM/YYYY; HH:MM), Stir Speed (the speed at which the solution was mixed, rpm), Settle time (the amount of time the solution rested before use in dilutions, hr), Notes (which cross of fish were in each exposure tank), Prepared by (referring to the scientists responsible for solutions preparation).
The worksheet “CEWAF Dilution Preparation”: Date and Time (referring to when dilution was made; DD/MM/YYYY; HH:MM), Carboy ID (referring to carboys used in transfer), Stock volume (amount of stock solution used to prepare dilution, L), Final volume (total amount of diluted CEWAF prepared, L), Prepared by (referring to the scientists responsible for solutions preparation).
The worksheet “Hatch and Survival": Cross (referring to which fish were spawned), Female Pit Tag ID and Males Pit Tag ID (identifying reference number for specific fish in spawning group), Spawning tank (referring to tank used for spawning event), mean total eggs (number of eggs counted from spawning event), mean total viable eggs (number of viable eggs counted from spawning event), mean fertilization (percentage of viable eggs fertilized in spawning event, %), mean hatch (percentage of fertilized eggs which hatched, %), mean 3-day larval survival (percentage of larvae which survived for 3 days post hatch, %).
