Abstract:

The current dataset is an extension of DOI:10.7266/BFXBPAD8 (Effects on the teleost kidney following early life-stages exposure to Deepwater Horizon crude oil). This dataset contains qPCR data, whole-mount immunohistochemistry images, and whole-mount in situ hybridization images.

Purpose:

(1) gain insight as to the likely underlying molecular mechanisms of crude oil toxicity on fish ELS pronephros development and function by assessing the transcriptional responses of candidate genes involved in these processes; (2) examine potential morphological defects induced by crude oil exposures during teleost pronephros development; (3) examine the acute and latent impacts of crude oil exposure during fish ELS on pronephros function by assessing osmoregulatory endpoints.

Methods:

Zebrafish tests: embryos/ larvae were exposed to 0% (control), 20% and 40% HEWAF solution from ~2 hours post fertilization (hpf) to 96 hpf. They were transferred in clean control medium for an additional 3 days, now 7 days post fertilization (dpf). During test M205b, embryos/ larvae were collected at 48 hpf, 96 hpf, and 7 days post fertilization (dpf) to assess transcriptional changes in genes involved in kidney development and function by qPCR. During test M211, samples were collected for whole-mount in situ hybridization analysis (looking at the transcripts with significant differences observed by qPCR) at 24, 48, 72, and 96 hpf, and for whole-mount immunohistochemistry at 72 hpf using an antibody raised against the Na⁺/K⁺-ATPase to observe the pronephros (early life stages kidney). Red drum test: during test E206, larvae were exposed to 0% (control) and 1% HEWAF solutions from 12 hpf to 36 hpf (24 h). Subsequentially, they were transferred into clean sea water up until 14 dpf for further studies. Larvae were collected at 36 hpf, 60 hpf, 84 hpf and 14 dpf to assess transcriptional changes in genes involved in kidney development and function by qPCR.
