Abstract:

Although aerobic capacity and swim performance have been relatively well-studied in the highly active pelagic Mahi-mahi (Coryphaena hippurus), less is known about what role adrenergic responses play in maintaining performance during periods of intense exercise or stress. The present dataset will contain measurements of maximum levels of adrenaline and noradrenaline in mahi blood immediately following a chase to exhaustion protocol.

Purpose:

To measure maximum adrenaline and noradrenaline in Mahi-mahi (Coryphaena hippurus) blood following a chase to exhaustion protocol.

Methods:

To measure maximum circulating adrenaline and noradrenaline in the blood of Mahi-mahi (Coryphaena hippurus). Adult Mahi-mahi (Coryphaena hippurus) were taken directly from rearing tanks and placed into an experimental chase tank and chased until they were exhausted. Following exhaustion, mahi were immediately euthanized with a blow to the head and a ventral incision was made to expose the heart. Blood was taken via cardiac puncture. Blood was collected using a 22-gauge blood collection needle attached to a 3.0 mL blood collection tube spray-coated with 5.4 mg K2 EDTA (dipotassium ethylenediaminetetraacetic acid, Vacutainer, BD Biosciences). Two tubes of blood were taken in rapid succession from each cardiac puncture. The first vacutainer tube was preserved with glutathione (5 microliters per 100 milliliters blood, 0.2 M), while no further preservative was added to the second vacutainer tube. Samples were stored at -80ºC prior to analysis. Samples were measured using Ultra-Performance Liquid Chromatography (UPLC) (ACQUITY model, Waters Corporation, Milford, MA) using fluorometric detection at 480 nanometer with excitation at 345 nanometer. Concentrations for adrenaline and noradrenaline are reported in micromolar.
