Abstract:

Recent transcriptomic studies on oil-exposed fish suggest effects on cholesterol homeostasis during early life stages. Cholesterol and biosynthetically downstream metabolites of cholesterol were measured following oil exposure in embryonic zebrafish (Danio rerio). Cholesterol was measured using a spectrophotometric assay, and downstream metabolites were measured by a contracted lipidomics core facility. In addition to lipid measurements, the dataset also includes water quality measurements and PAH measurements and supporting data such as how HEWAF (high energy water accommodated fraction) was made.

Purpose:

Understand effects of oil on cholesterol and downstream metabolites in developing teleosts, specifically zebrafish (Danio rerio).

Folder Lipidomics study contains the following files: Lipid Panel; Lipidomics Metadata; Sample Inventory; Tank ID & Stock Code; Test Conditions Table; Test Performance Monitoring; TPAG Analysis K2001809; WAF Preparation; Water Quality Monitoring. Each file contains multiple headers including the following: Treatment; Age; Survival; pH; dissolved oxygen (mg/L); PAH concentrations (µg/L)

Methods:

High energy water accommodated fractions (HEWAFs) of OFS oil were prepared daily at a concentration of 1 mg/L of oil to water and dilutions were made accordingly to 40%, 20%, 10%, or 0% of HEWAF. Test duration was 96 hours, where mortality and water parameters (pH, temperature, dissolved oxygen) were measured daily for each replicate. Zebrafish (Danio rerio) were used, and exposure started ~4 hours post fertilization (hpf) and lasted to 96 hpf. Animals were collected at 48 hpf and 96 hpf. These samples were sent to VCU lipidomics core for a sterol panel analysis.
