Abstract:

Dataset contains measurements of transendothelial electrical resistance of mouse brain microvascular endothelial cells after exposure to controls and test compounds: crude oil high energy water accommodated fraction (HEWAF), benzo[a] pyrene, and Corexit. Purpose:

To study the effects of benzo(a)pyrene (BaP), COREXIT, or HEWAF on mouse brain microvascular endothelial cells.

Data Parameters and Units:

The dataset contains 3 excel files, “ECIS_Data.xlsx”, “MTT_Assay.xlsx”, and “PCR.xlsx”. The file "ECIS_Data.xlsx" contains data on transendothelial electrical resistance, the file "MTT_Assay.xlsx" contains the makeup of the various treatments, and the file PCR.xlsx contains data on gene expression. Benzo[a]pyrene = BAP; High energy water accommodated fractions = HEWAFs; A550-A690; Gapdh = glyceraldehyde 3- phosphate dehydrogenase; claudin-5 = Cldn5; vascular cell adhesion molecule-1 = Vcam1; Tight junction-associated protein =Tjp1; and occludin = Ocln.

Methods:

High energy water accommodated fractions (HEWAFs) were prepared from “source B” crude oil, as described previously. 200 mg of source B” crude oil was mixed with 1 l of complete EC media. The suspension was mixed for a duration of 2 mins. This was followed by vortexing at high speed for 5 min. after settling for 30 mins 80% of the lower solution was siphoned out. This solution represented 100% HEWAF which was diluted.

Instruments:

The Electric Cell-substrate Impedance Sensing (ECIS) system (Applied BioPhysics, Troy, NY). Was used to measure transendothelial electrical resistance (TEER; Ω∙cm2 ) at 4000 Hz.
