Abstract:

The present dataset is of assays of the testing the role of exposure to Polycyclic Aromatic Hydrocarbons (PAHs) via vapors or via diet on transgenerational epigenetics in the king quail (Coturnix chinensis). Exposures were performed via diet on the parental generation. Endpoints for this experiment for the parental population are estimation and comparison of body mass, organ mass, and oxygen consumption among experimental treatments. For the offspring generation obtained from the parental groups we measured morphological characteristics of the eggshells (mass, length, and width) and physiological characteristics such as egg water loss, number of pores per area, and animal oxygen consumption. This data set contains images of the eggshells and the raw data files of oxygen consumption.

Purpose:

To determine if dietary exposure to crude oil leads to effects in the offspring population, from tissues to the organism.

Methods:

1000 mg of crude oil were added into 1L of conditioned aquarium water and blended for 30 seconds in a commercial blender (WaringTM CB15). After blending, the mixture was placed into a separation funnel for 1 hour, after which 100 ml of the solution was taken out through a bottom port of the funnel and discarded. 600 ml of the remaining mixture (considered as 100% HEWAF) and two diluted solutions (1% and 10% HEWAF in deionized water) were used for diet preparation. Adult king quails (Coturnix chinensis) were separated in 5 groups composed of 5 breeding pairs. Each group was exposed to control food, low crude oil concentration diet, or high crude oil concentration diet during at least 3 weeks prior breeding. Body mass was measured weekly, metabolic rate was estimated by flow-through respirometry. Fecundity per experimental group each other day was determined by direct counting of the number of eggs laid per female. The eggs were measured and weighed. Water loss through the eggshell was measured each other day in newly hatched eggs. The number of pores and their size in the eggshells were estimated by digital analysis in the software ImageJ. Hatchling success was estimated per experimental group. Body mass of the chicks was estimated at hatching. Summit metabolic rate was estimated by flow-through respirometry. Hematological variables were estimated by obtaining blood directly from the artery.
