Abstract:

Surface seawater from the Aransas Channel, Port Aransas, Texas was incubated with crude oil at initial concentrations of 0 ppm, 50 ppm, 100 ppm, 200 ppm, 400 ppm, and 800 ppm. The samples were incubated at room temperature for 50 days. Bacterial composition was sampled and analyzed at 0, 5, 12, 28, and 50 days. This dataset represents the bacteria composition at genus level for different concentrations and various time points.

Purpose:

To determine the relative abundances of bacteria when seawater was amended with different concentrations of oil.

Methods:

Surface water (~1 m) was collected in July 2013 from the University of Texas Marine Science Institute Pier Lab located in the Port Aransas Ship Channel, Texas (27.84°N, 97.05°W). A portion of the sample (~1 L) was filtered through 0.2 μm nylon filter to remove particles and microorganisms including bacteria. Using serological pipette, 20 mL of the filtered water was transferred into a 50-mL pre-combusted amber bottle. Subsequently, 2.5 mL of the unfiltered seawater as inoculum was added to each bottle. Light Louisiana sweet crude was added to the bottles to attain final concentrations of 50 ppm, 100 ppm, 200 ppm, 400 ppm and 800 ppm. Bottles containing no oil were prepared as control (0 ppm). The oil was added directly to the bottle using a sterile pipette tip. DNA was extracted, followed by PCR amplification and DNA sequencing was done with 454 Pyrosequencing.
