Abstract:

A number of bacterial species have been identified that can consume petroleum hydrocarbons with diverse metabolic capabilities. Laboratory experiments were conducted to investigate bacterial consumption by monitoring the volume change of oil droplets as well as effects of oil droplet size on this process. Two purified exopolymeric substances (EPS) from bacterial consortia were used. This dataset contains both time lapse and high-speed micrographs of EPS purified from bacterial consortia.

Purpose:

To determine the change in crude oil microdroplets morphology and EPS aggregation using purified EPS of microbial consortia.

The folder "Consortia_R6_0011_R6.x802.0000011" contains two subfolders as follows: 1. 1_20_2018_experiment size=28.5 GB; Contains 2 subfolders on time-lapse images in tif files, and 7 videos in nd2 format. 2. 8_27_2018_experiment size=19.7 GB; Contains 2 subfolders on time-lapse images in tif files, and 4 videos in nd2 format

Methods:

The experiments were conducted using the new ecology-on-a-chip (eChip) microfluidic platform. The eChip is a platform that enables us to study microbial interactions with a rising droplet at ecologically relevant length and time scales. This method generates a micro-scale droplet in a microfluidic channel that allows suspended bacteria to naturally flow and interact with the droplet simulating a real environment.
