Abstract:

Soil samples were collected from 21 designated stations in northern Barataria Bay, which included 7 reference, 7 moderately oiled, and 7 heavily oiled stations. For the soil microbial community, DNA was extracted from soil samples, sequenced with Illumina MiSeq, and the sequence data was processed with QIIME.

Purpose:

To examine responses of soil microbial community structure and function to the various oiling intensities. Multi-year sampling is intended to evaluate the recovery of the soil microbial community.

Methods:

Samples were collected twice a year from 21 designated sampling sites that included 7 heavily oiled, 7 moderately oiled, and 7 reference sites. Heavily oiled marshes had a thick coating of viscous, emulsified oil that completely covered plant shoots, most of which were laid down horizontally, and coated the soil surface; the average TPH concentration was 511 ± 231 mg g−1 dry soil in January 2011. Moderately oiled marshes generally had partial oil coverage on the lower portion of plant shoots, most of which were able to maintain a vertical position, in combination with intermediate soil-oil concentrations in the soil; the average TPH concentration was 70±38mg g−1 dry soil in January 2011. The top two cm of soil was collected at each site, proximal to the site of collection of the other types samples from this study. The soil was transported to the lab on ice and stored at -80°C. DNA was extracted from each soil sample, checked for purity, and amplified by PCR of the V4 barcode region, 16S rDNA for prokaryotes. Sequencing of PCR amplicons was conducted with Illumina MiSeq. DNA processing was done in QIIME v. 1.9.1 with default settings except for a Phred score of 25. Please contact the authors for further information.
