Abstract:

This dataset includes microsatellite data from different populations of adult Tabanus nigrovittatus sampled in coastal Louisiana in 2015 and 2016. The 2015 dataset is based on ten microsatellite loci; the 2016 dataset contains eleven loci.

Purpose:

A population genetics study of the recovery of Tabanus nigrovittatus populations five to six years after the oil spill.

Data Parameters and Units:

Microsatellite genotypes consist of two alleles identified by their 3 digit base pair length per locus. Missing data are encoded by -9.

Methods:

Total DNA was extracted from thoraces of up to 30 adult female horse flies per sample (trap) using the DNeasy Tissue Kit (Qiagen Inc., Valencia, CA) and genotyped using the Li-Cor 4300 automated DNA analyzer (Li-Cor Inc., Lincoln, NE) as described in: Husseneder, C., Delatte, J.R., Krumholt, J., & Foil, L.D. 2014. The same microsatellite loci developed, rigorously screened and used in previous population genetic studies of the greenhead horse fly (Husseneder et al. 2014) were employed to compare population genetic parameters in 2015 and 2016 to those measured following the population crash of this species immediately after the oil spill (2010, 2011) published in: Husseneder, C., Donaldson, J.R., & Foil, L.D. 2016. Since a formerly monomorphic locus (7FA, Husseneder et al. 2014) showed multiple alleles in several samples of 2016, the number of polymorphic loci was increased from 10 (2010, 2011, 2015) to 11 (2016). Locus characteristics, GenBank accession numbers, and other summary statistics have been published previously (Husseneder et al. 2014, 2016).
