Abstract:

As part of a study to investigate the effect of natural seepage on deep-sea corals, the metagenomes of the octocoral, Callogorgia delta, and the surrounding sediment were sequenced. At sites within lease blocks MC751 and MC885, Callogorgia delta colonies can be found growing near visual signs of active seepage. Colonies were collected on 2015-05-02, 2016-10-01, 2016-09-29, 2017-06-06, and 2017-06-18 growing near or far from signs of seepage. Sediment samples were collected near corals with push cores. The metagenomes were sequenced to investigate the effects of natural seepage on the composition of the microbial communities associated with corals and sediment and to determine any changes in metabolic capabilities in these microbial communities.

Methods:

DNA was extracted from coral samples using a DNA/RNA allprep kit and DNA was extracted from sediment using a Qiagen powersoil kit. DNA extracts were sequenced on an Illumina Hiseq 2500 platform using 100 bp paired end reads.
