Lipid class composition of red drum fed a diet of pellets saturated with south Louisiana crude oil using Iatroscan.

Purpose:

Controlled oil exposure study to evaluate the effects of crude oil on lipid structure in whole body of fish.

Data Parameters and Units:

Food Preparation worksheet: Prep start and end date, est. total mass needed for daily feeding and samples (g), feed prep. method, feed mass (g), mass of oil added (g), mixing speed, mixing time, oil source. % Lipid worksheet: FISH ID, Tank ID, Whole Body (sample ID), % Lipid, Liver (sample ID), % Lipid. Liver TLC Results worksheet: Fish ID, Tank ID, REP (replicate), Liver MML #, % Wax esters & Sterol Esters, % Triglycerides, % Free Fatty Acids, % Cholesterol, % Phospholipids & Polar Lipids. Methods:

Tissue samples extracted via EPA method 3545 using a Dionex ASE300, then concentrated to 10 mL in DCM for analysis. Lipid Composition: 1 uL of sample analyzed with Iatroscan using PeakSimple 444 software.

Error Analysis:

QA measures were included into per batch according to Environmental Laboratory for Forensics Quality Assurance Plan. If QA measurements were not met, samples were re-analyzed or data was qualified.
