Abstract:

As a part of CARTHE’s SPLASH (Submesoscale Processes and Lagrangian Analysis on the SHelf) experiment, sea surface microlayer and subsurface samples were collected on polycarbonate membrane filters in the Louisiana Bight, northern Gulf of Mexico on 19-22 April, and 25 April 2018. This dataset contains calculations of absolute abundance from Illumina MiSeq data for surfactant-associated bacteria. Absolute abundance results of surfactant-associated bacteria were obtained via sequencing (Illumina MiSeq) from Argonne National Laboratory and analysis was performed using QIIME (Quantitative Insights Into Microbial Ecology). Also, confidence intervals were calculated using the negative binomial method.

Purpose:

To determine the bacterial microlayer composition off the Louisiana coast.

Data Parameters and Units:

OTU_File.xlsx: Absolute bacterial abundance (OTU counts) for all samples and all bacteria; SampleID: sample number; following headers are bacterial genera, starting with Unassigned. Genera_Avgs.xlsx: Average bacterial genera abundance for each site (OTU); Site (A-G); Type (SML-microlayer, SSW-subsurface water, NE-non-exposed controls); following headers are genera of bacteria, other (non-surfactant associated and unassigned), and last column is total count of OTUs including all genera. CIs.xlsx: calculation of average number of OTU hits and confidence interval using the negative binomial formula for genera Halmonas, Oleispira, and Alcanivorax; SML or SSW: sea surface microlayer or subsurface water samples used in calculation; Site: A-G, Controls (NE: non-exposed, AC: air control, and NTC: non-template control; OTU Count: average count of OTU hits at each site for that type; T: Student’s distribution coefficient for 95%; T/2: Student’s distribution coefficient divided by two for use in negative binomial formula; STDEVPA: standard deviation; N: number of samples; MIN_CI: minimum confidence interval calculated using the negative binomial formula; MAC_CI: MAX_CI= maximum confidence interval calculated using the negative binomial formula. The samples were collected from sites A-G. For each site, there is a beginning and end coordinate. Methods:

Samples were collected from a small boat on April 19-22, and April 25, 2018 in the Louisiana Bight, northern Gulf of Mexico.
