Abstract:

This dataset contains marine aggregate stain pictures from mesocosm treatments of seawater control, WAF (water accommodated fraction of Macondo oil), CEWAF (chemically enhanced WAF through adding Corexit), and DCEWAF (diluted CEWAF) using a Fluorescence Microscope during the period of August-October 2015. Briefly, different dyes were applied to sediments to highlight different components and capture the size and intensity variations between treatments.

Purpose:

To enhance the visibility of compositional changes sediment marine aggregates under different treatments. These data show that the formation and composition of aggregates changed when oil and dispersant were introduced into water column.

Methods:

Nile red, Concanavalin A-FITC, and Chlortetracycline hydrochloride were used to stain hydrophobic, polysaccharides, and DOM components, respectively, in sediment aggregates in samples from different mesocosms. Samples of sediment were mixed with 1:200 Nile red, Concanavalin A-FTIC, and Chlortetracycline hydrochloride. The fluorescence measurements were obtained by Nikon microscope (Nikon Eclipse TE2000- U, Tokyo, Japan). Pix number was sorted by image J and redrawn by Prism 6, each line indicated the average of three independent pictures. Nile is a hydrophobic dye (Molecular Probes, NY, USA); Concanavalin A conjugated FITC (Sigma-Aldrich, USA) is a carbohydrate binding protein that stains polysaccharides. Chlortetracycline hydrochloride (Sigma-Aldrich, USA) is a BioReagent suitable for staining divalent calcium cations.
