Abstract:

These data are of marine microgel formation from EPS (extracellular polymeric substances) harvested from 4 different phytoplankton and the effect of oil and Corexit impact on their formation. The data collected are from DLS (dynamic laser scattering) as the gels form over time.

Purpose:

To measure the microgel aggregation behavior when Corexit and oil introduced into marine ecosystem.

Methods:

The size of microgel was monitored by dynamic laser scattering (DLS) following the published protocol. Briefly, samples were shaken, and filtered through a 0.22-μm Millipore membrane (pre-washed with 0.1N HCl) before use. Aliquots were then poured directly into scattering cells. The scattering cells were positioned in the goniometer of a Brookhaven laser spectrometer (Brookhaven Instruments, NY, USA). The polymer assembly was monitored for 8 ~ 10 days by analyzing the scattering fluctuations detected at a 45 degree scattering angle. The autocorrelation function of the scattering intensity fluctuations was averaged over a 10-min sampling time, using a Brookhaven BI 9000AT auto correlator. Particle size distribution was calculated by the CONTIN method. Each measurement was taken in triplicate in 10 ml at room temperature. A calibration of the DLS method was conducted using standard suspensions of latex microspheres (Polysciences, PA, USA). For this research, seawater was purchased from Bigelow Lab for Ocean Sciences. The four phytoplankton used are: Dunaliella tertiolecta (UTEX999) Thalassiosira psaeudonana (TP) Ampora sp. Sagittula stellata EPS was provided by Zhang and Santschi and purified using the protocol in Zhang and Santschi, 2009. EPS concentration is 100 ng/L in seawater and with 1ppm of WAF, CEWAF or Corexit. Protocol of how to make WAF and CEWAF: WAF: 1. 1L filtered seawater (0.2um twice) 2. Pasterized 65C for 4hrs. 3. 200 ul crew oil add into seawater, mix slow for 24hrs at room temperature. CEWAF: 1. 1L filtered seawater (0.2um twice) 2. Pasterized 65C for 4hrs. 3. Mix 10ml crew oil and 500ul Corexit room temperature for 24 hrs. 4. Add step 3 20ul into step 2 seawater. Both of them final carbon content will be 4mg/L. Last step is filter sample by 0.22um filter before put into DLS vial. Each vial is 10ml. Repeat six times, thus S1 to S6 are replicate samples run in the DLS.
