Abstract:

This dataset reports how oil concentrations affect oil degradation rates and the development of bacterial community through laboratory incubations. Alkane analysis, bacterial cell density, and bacterial community structure are included.

Purpose:

To examine how oil concentrations affect oil degradation rates and the development of bacterial community.

Data Parameters and Units:

Alkanes worksheet: alkanes analysis was conducted at 50 ppm, 400 ppm, and 800 ppm for days 0, 5, 12, 28, and 50. Values are the average of three replicates in mg/L, and "d" after the number represents days of incubation. Bacterial cell density worksheet: Bacterial cell density was measured using flow cytometer (Accuri C6) after the samples were dyed with SYBR Green I. Note: the two exact sample names represent duplicate analysis. T0 represent time zero day. Ship Channel surface water is the original bacterial abundance in the ship channel water. Sample name, Bacterial count (30uL), Cells/mL. Bacterial community structure worksheet: bacterial community were pyrosequenced using Eubacterial primers 28F 5′TTTGATCNTGGCTCAG-3′ and 519R 5′GTNTTACNGCGGCKGCTG-3′ to amplify the 500 bp region of 16S rRNA gene. Data presented here are in genus level.

Methods:

Seawater was collected from Deepwater Horizon site (2m) in May 2013 and Ship Channel of Port Aransas in February 2014. The water from DWH was filtered through 0.2um, and 20.5ml was added into 30 mL pre-combusted amber bottles. 2.5mL unfiltered ship channel was added as inoculum. Louisiana light sweet crude oil (BP) was added to the glass vials to make the final oil concentrations of 0, 50, 100, 200, 400 and 800ppm. There were 5 glass bottles at each time point, and three were used for oil analysis (n-alkanes and PAHs), one for nutrient analysis (ammonium, nitrite+nitrate, phosphate), and one for bacterial abundance and community. The incubation experiment lasted 50 days, with sampling at 0, 5, 12, 28, and 50 days.
