Abstract:

This data indicates the growth change of dinoflagellate Karenia brevis after the addition of altered bacterial communities exposed to crude oil and light. To investigate the effect of bacterial communities on the initiation phytoplankton bloom in oil-polluted environments, (i) we exposed bacterial communities to crude oil (200 ppm) and light, (ii) inoculated these altered bacterial communities into dinoflagellate K. brevis, (iii) and then measured the growth change of K. brevis and bacterial density. This experiment was conducted in the laboratory and data was collected from October, 2016 to February, 2017.

Purpose:

The cell density of Karenia brevis and bacteria, Time (days), Growth promoting effect (%), Average (cells/mL), Cell concentration, Standard deviation, Volume (uL) The effect of oil and light-Phy worksheet: To investigate the effect of altered bacterial community structure exposed to crude oil and light, I exposed two bacterial community structure which are free-living (FL) and particle-associated (PA) bacteria to crude oil under the presence (light) and absence (dark) of light conditions. Thus, FL+Oil (light) indicate the treatment of the addition of free-living (FL) bacterial community structure exposed to crude oil and light. The oil concentration in this experiment was 200 ppm. Experiments were conducted in duplicate, labeled I and II. Raw data: the actual counting number of Karenia brevis (dinoflagellate) via light microscopy. Direct counting was also carried out in duplicate (I and II). Invert to per mL: Convert the actual counting number to "cells per milliliter". Final: The average density (cell per milliliter) of K. brevis in control and each treatments. Ave and Stdev indicate average and standard deviation, respectively. Growth promoting effect (%): To figure out the growth promoting effect, this value was calculated by following equation: (cell density of Treatment - cell density of control) / (cell density of control) x 100 The effect of oil and light-Bac worksheet: bacterial densities of control and each treatments (same experiment to the first sheet). The effect depending on density worksheet: To determine whether this altered bacterial community structure enable to stimulate the growth of K. brevis or not, I inoculated two different densities of altered bacterial community structure (higher and lower). Higher and lower concentration was constructed by harvesting 35- and 15 ml of treatments which are FL (free-living) and FL+PA (free-living and particle associated) bacteria exposed to crude oil and light. Raw data, invert to per mL, final, and growth promoting effect (%) indicate same things with the first sheet. Positive control-Phy worksheet: In this study, bacteria which are established in the xenic culture of K. brevis were used. This implies that these bacteria may be closely associated with host dinoflagellate growth. Then, it is unclear whether the growth promoting effect is the result of altered BCS exposed to crude oil and light or the addition of associated BCS, regardless of treating crude oil and light. To resolve this, the effect of FL BCS which isolated from the stationary phase of K. brevis culture on the growth of host dinoflagellate was investigated as positive control.
