Abstract:

This data presents the results of the RNA sequencing of Psedomonas aeruginosa (PAO1 & PA14) cells confined at the hexadecane-water interfaces vs. control cells (in the saline solution). RNA sequencing was performed to characterize the change in the transcriptional profile of the cells after interfacial confinement. Also, live/dead imaging was performed to evaluate the physiological stress induced by the confinement at the oil-water interface, known as FBI (films of bacteria at interfaces) conditions. Related video files and growth dynamic data can be found in GRIIDC datasets R4.x259.000:0004 and R4.x259.000:0003, respectively. Purpose:

To sequence the RNA of two Pseudomonas aeruginosa strains (PAO1 and PA14) after exposure to hexadecane-water interfaces to established their transcriptomic profiling.

C1 is for the green channel and C2 the red. For instance: C1-PA14 FBI and PAo1 D1.lif - Series016 indicate green channel (Live cells) C2-PA14 FBI and PAo1 D1.lif - Series016 indicate red channel (dead cells) PA14 FBI and PAo1 D1.lif - Series016-1 indicate combined image merging of the C1 and C2 or green and red channels. Adhesion time indicated in file name as either 30 minutes or 1 hour. In the folder PAO FBI sample 1: The .avi file is a Z-stack imaging of one of the oil emulsion covered by P. aeruginosa cells. It shows live cells adhering to the interface.

Methods:

Pseudomonas aeruginosa PAO1 and PA14 cells were used. For this experiment the cells were diluted to 154 nM sodium chloride solution to reach an optical density of 0.2 at 600nm. After 1 hour in this solution cells from both strains were sampled and total RNA was isolated as a control. To form films at bacteria interfaces (FBI) 5 mL of the suspension of cells in NaCl and 5 mL of hexadecane were mixed in 25 mL glass vial by rotating using a Rugged Rotator (60 rpm for 1 hour). The resulting emulsion was collected and centrifuged (15, 000 g for 2.5 min) to remove cells. FBIs were collected from the hexadecane water interfaces. RNA was extracted using RNeasy Mini Kit. RNA samples were sent to the Penn Next-Generation Sequencing Core for sequencing.
