Abstract:

This dataset contains video files of the microfabrication of microbial nanocultures, as well as the growth dynamics of the encapsulated cells. Pseudomonas aeruginosa cells were encapsulated in polydimethylsiloxane (PDMS) membrane nanocultures. Video data show stained cells, long-term encapsulation, the process of generation of the nanocultures using a microdevice, osmotic annealing of the nanocultures, and growth within the nanocultures. This dataset is related to dataset R4.x259.000:0003.

Purpose:

To assess the cultivability of microbes using nanocultures in order to study microbial persistence.

Data Parameters and Units:

Live dead Staining od PAO1 NC.mp4: P. aeruginosa PAO1 was encapsulated in polydimethylsiloxane (PDMS) nanoculture overnight and the nanocultures were stained with Filmtracer (life technologies), comprising Syto 9 (green stain) and propidium iodide (red stain). Long term encapsulation.avi: The PDMS nanocultures were incubated in 154mM NaCl solution for 7 days and 7 months. During that period of time, the nanocultures were imaged to assessed the integrity of the nanocultures, as well as the viability of the encapsulated cells. micro_device.avi: We make use of a glass-capillary microfluidic device with hydrodynamic flow-focusing and coflowing geometry to generate the W/O/W PDMS capsules. This video show the process of generation of the nanoculture. The flow rates of the inner, middle and outer phase were set to 300 μL/h, 750 μL/h, and 10 mL/h, respectively. Osmotic gradient_PDMS NC 5h video.avi: We impose a strong osmotic stress (10% NaCl) for 5h around nanocultures that had been polymerized after 24h of incubation at 37°C resulted in wrinkling and collapse of the PDMS nanocultures. Growth of PAO1 in PDMS for 16 h each 50th frame is 500s.mov: The growth kinetics of P. aeruginosa PAO1 in PDMS nanoculture. Nanocultures containing P. aeruginosa PAO1 cells in UFTYE medium were collected in 154mM NaCl solution and cell growth was monitored at 37°C for 16 hr using a microscope.
