Abstract:

The aim of this study was to investigate the combined effect of oil from the surface (OFS) high energy water accommodated fraction (HEWAF) exposure and chronic temperature (26 and 30°C) on heart developing of mahi-mahi. Five HEWAF concentrations were tested, 0.5%, 1%, 2%, 4% and 8% OFS HEWAF (100%=oil loading at 1g/L). Viable eggs were exposed 24 hours and then transferred in clean sea water until 48 hpf. Cardiac assessment (heart rate, stroke volume, cardiac output, cardiac morphology) were made in hatched-larvae (48h).

Purpose:

To examine the morphology and cardiac function changes following oil exposure combined to two chronic temperatures exposures in early-life stages of mahi-mahi.

Methods:

All data were collected from image and video acquisition. Morphometric measurement were made using Image J software with the freehand tool. Stroke volume is determined by the difference between the end-diastolic volume and the end-systolic volumes. Volumes are determined by the prolate spheroid equation. Cardiac output is the product between stroke volume and heart rate.

Instruments:

Nikon SMZ800 stereomicroscope coupled to a Fire-i400 or Fire-i530c digital camera (Unibrain, CA) + thermal microscope stage temperature controller (Book Industries, IL)
