Abstract:

The timing and location of the 2010 Deepwater Horizon (DWH) incident within the Gulf of Mexico pelagic zone likely resulted in exposure of commercially and ecologically important fish species, such as mahi-mahi (Coryphaena hippurus) during the sensitive early life stages. A 24-channel optical fluorescence oxygen sensing system for high through-put respiration measurements was used to investigate the individual and combined effects of oil exposure, temperature and developmental stage on oxygen consumption rates in embryonic and larval mahi-mahi. This dataset reports oxygen concentrations that can be used to calculate embryonic oxygen consumption (picomol inidiv-1 min-1) rates for 2 temperature treatments and 3 oil concentration treatments for day 1 (0-24 hr), day 2 (24 -48 hr) post-fertilization (hpf) mahi-mahi embryos and for recently hatched larvae (~50 hpf). Nitrogenous waste data is also reported for larvae exposed to different concentrations of oil and different temperature treatments. This data is reported as ammonia, urea and total nitrogenous waste excretion (picomol inidiv-1 min-1).

Purpose:

Testing effects of different concentrations of oil exposure on oxygen consumption, energy utilization and cardiac abnormalities in embryos and larvae of mahi-mahi.

Data Parameters and Units:

Ammonia, urea and total nitrogenous waste excretion in mahi-mahi embryos and larvae (pmol indivi-1 min-1)- data found in ‘NW Complied Data’ folder. The first 3-5 tabs include the raw data from the different treatment conditions (temperature oC and % HEWAF dilution) and the last 3 tabs have the data corrected for controls and expressed as (pmol indivi-1 min-1). Oxygen Consumption (% air saturation and umol L-1)- data found in ‘Microplate Raw Data’ folder within additional folder labeled by the different treatment conditions (temperature oC and % HEWAF dilution). Data is presented as either % air saturation or umol L-1 by elapsed time (seconds). Images and videos taken to measure heart rate, pericardial and yolk sac edema and yolk sac depletion- data found in ‘Scope A’ and ‘Scope B’ folders. Specific calibration files for each scope found in separate folder named ‘Calibration Pics and Notes’- Files are named by measurement unit and by the magnification set on microscope. The notes document provides information on the magnification used for pictures and videos. Sum PAH data- found in folder titled ‘Sum PAH data’. Raw data provided a long with break down and percentages of different ringed PAHs. Methods:

Surface slick oil from the DWH spill was prepared as a high-energy-water-accommodated fraction (HEWAF) to generate environmentally relevant PAH concentrations and compositional profiles. Loligo Microplate was used to measure oxygen consumption in developing mahi-mahi. Micro modified colorimetric assays were used to measure ammonia and urea excretion.

Instruments:

Oxygen Consumption- Loligo Microplate (24 well optical-fluorescence, oxygen-sensing system). Images and videos were taken using either a Fire-i400 or Fire-i530c digital camera (Unibrain, San Ramon, CA) mounted on a Nikon SMZ800 stereomicroscope.
