Abstract:

The aim of this study was to investigate the effects of oil naturally irradiated on the survival, whole morphology and cardiac function in larval red drum (Sciaenops ocellatus). A range of concentrations of irradiated Source Oil B (SOB) were tested. Morphological and cardiac variables were assessed during the first developing 48 hours.

Purpose:

To examine the morphology and cardiac function changes following oil exposure combined to two chronic temperatures exposures in early-life stages of red drum.

The dataset contains 6 excel files which contain information on the chemistry of the medium, heart measurement, heart rate measurement, standard length of fish, survival, ultraviolet data and weather-temperature data. Dataset also contains 2 folders of videos and pictures, and text file explaining the naming convention of pictures and videos. Methods:

The high energy water accommodated fractions (HEWAFs) were generated at ambient temperature and prepared at a loading rate of 1 g of oil (Source Oil B) per liter of UV-sterilized seawater (35 ppt) and mixed in a Waring CB15 blender (Torrington, CT) at low speed for 30 s. Immediately after mixing, the solution was transferred to a glass separatory funnel for 1 h in the dark. After settling, the 90% lower portion of the solution was carefully drained and retained for subsequent use as 100% HEWAF. This solution was diluted to 60% with ambient seawater for the solar radiation process. UV radiation of HEWAF was by natural sunlight. Red drums were then exposed to different concentrations of SOB under laboratory conditions. All data were collected from image and video acquisition. Morphometric measurements were made using Image J software with the freehand tool. Stroke volume is determined by the difference between the end-diastolic volume and the end-systolic volumes. Volumes are determined by the prolate spheroid equation. Cardiac output is the product between stroke volume and heart rate. Total PAH characterization and quantification have been made by ALS Environmental (Kelso, WA, USA), according to standards of Deepwater Horizon Natural Resource Damage Assessment toxicity testing program.

Instruments:

Biospherical radiometer (BioSpherical Instruments, San Diego, CA) - Nikon SMZ800N stereomicroscope connected to a Nikon Digital DS U-3 camera.
