Purpose:

This research provides a set of techniques for visualization and analysis of three-dimensional (3D) microtissues. These methods have been optimized for toxicological assessment of 3D models and specifically address the challenge of visualizing the internal features of microtissues.

Data Parameters and Units:

*.LSM Confocal image files of BEAS-2B human lung epithelium, LNCaP human prostate adenocarcinoma, PLHC-1 Poecilopsis lucida hepatocyte both cleared using ClearT2 protocol and uncleared

Methods:

Images were obtained using a Zeiss LSM710 laser confocal microscope and a 20x objective (Zeiss LSM7110, Plan-Apochromat, 0.8 NA, 0.55 WD objective). Images can be viewed using Zen lite software from Zeiss at https://www.zeiss.com/microscopy/us/products/microscope-software/zen-lite.html. Tissues were cleared using a modified version of the ClearT2 protocol described in Kuwajima et al 2013, ClearT: a detergent and solvent-free clearing method for neuronal and non-neuronal tissue, Development 140:1364-1368 doi: 10.1242/dev.091844.
