Abstract:

We have conducted systematic studies using a microfluidic platform to investigate the degradation rate of crude oil via physical and biological processes including oil dissolution, evaporation, and bio-consumption by hydro-carbon degrading bacteria. In the current study, we focused on the volume change of micro-oil droplet and structure of biofilm at oil water interface. The volume was measured using Atomic Force Microscopy (AFM) in Tapping mode. The biofilm structure or raft are measured using conventional inverted microscope. This dataset reports AFM data from experiments printing different sizes of base oil droplets (5, 10, 20, 50 micron) with pico-liter volumes in both square and circles. Results from patterning are also provided.

Purpose:

Systematic studies using a microfluidic platform were conducted to investigate the degradation rate of crude oil via physical and biological processes including oil dissolution, evaporation, and bio-consumption by hydro-carbon degrading bacteria.

Data Parameters and Units:

Three columns as x, y and height of the surface. All units are in meters.

Methods:

Volume measurement is provided by Atomic Force Microscopy (AFM). 3 Structure of biofilms are provided as micrographs. Surface profiles were obtained using Atomic Force Microscopy. Surface structures were visualized with microscope images. AFM measurement is obtained at temporal rate of 24 hours over a period of 7 days. The sample droplets were randomly chosen for at lease three droplets / sample. The micrograph measurement were conducted every 24 hours over 5 day period. Two different bacterial species, Alcarnivorex and Pseudomonas sp (PS62) were generated. AFM data files (.001, .002, .003, etc.) can be opened with free open source software Gwyddion 2.44. Instruments:

Burke Cube2000 AFM. Micrograph is obtained using Ti2000 Nikon Microscope with 20X and 40X Superplan ELWD objectives.
