Abstract:

This dataset reports the bacterial community structure of surface oil mousse, ambient surface water, bottom water, and sediment adjacent to the wellhead. Ambient surface water and surface oil mousse samples were collected in May 2010. Samples were collected from an oil spill site and a control site. Oil mousse samples were also collected from a marsh site July 21, 2010. Sediment core and grab samples were collected in May 2011 adjacent to the Deepwater Horizon wellhead. Water samples were also collected just above the core and grab samples. DNA was extracted at the Research and Testing Laboratory (Lubbock, TX) and bacterial tag-encoded FLX amplicon pyrosequencing (bTEFAP) was used to amplify the 500 bp region of the 16S rRNA gene.

Methods:

Surface water sampling: 2-2.5 L of surface water was collected by bucket, filtered using 47 mm 0.7um GF/F filter - glass filtration system. Oil mousse sampling: collected by bucket, transferred into polyethylene bottles (at Marsh Point, mousse was collected from marsh scrapings, mousse was emulsified with 40% water mass. Sediment sampling: Station grab - Ponar grab sampler, glass jar. Station core - sediment corer, section 0-2 cm. Approximately 250 mL of water above the sediment samples was also collected (about 5cm above the sediment). These samples were filtered (0.2um, 47 mm diameter Poretics polycarbonate filters). Filters and mousse samples were stored at -20°C until analysis.
