Abstract:

This dataset was generated to evaluate short-term changes in bacterial diversity from beach sand impacted by tides. The study was conducted on Dauphin Island, Alabama. The beach was oiled during the 2010 Deepwater Horizon oil spill, but the sand was not remediated by sand-washing. Sediment samples were intensively collected from twenty transects perpendicular to the shoreline (subtidal to berm) over a five day period in May 2012. Water samples were also collected, corresponding with each transect, and filtered through 0.22um Sterivex filters for DNA extraction. From extracted DNA, the V1-V3 region of the bacterial 16S rRNA gene was amplified, and single-direction reads were generated using multiplexed, bar-coded primers for pyrosequencing. Field data for all twenty transects and classified bacterial read inventories for four transects are available in corresponding dataset R1.x139.143:0003.

Purpose:

This dataset was generated to evaluate short-term changes in intertidal microbial communities on exposed sandy beaches over tidal cycles.

Data Parameters and Units:

The data are amplicon pyrosequence libraries of single-direction reads generated using multiplexed, bar-coded primers targeting the V1-V3 region of the bacterial of 16S rRNA gene.

Methods:

Total nucleic acids were extracted in triplicate. Three to six grams of beach sediment were subjected to DNA isolation methods that incorporated sucrose lysis buffer with lysozyme (1 mg/ml) and a solution of 5X proteinase K/CTAB/SDS; incubation at 55 degrees C while shaking at 40-120 rpm; nucleic acids precipitation in isopropanol at -20 degrees C; and ethanol washing. This approach was modified from Guerry et al. (1973), Somerville et al. (1989), Zhou et al. (1996), and Mitchell and Takacs-Vesbach (2008).

Instruments:

Amplification and pyrosequencing of the V1-V3 region of 16S rRNA genes were done using a 454 GS FLX Titanium (Roche, 454 Life Sciences, Branford, CT, USA) by the Molecular Research LP (Sweetwater, TX).
