Abstract:

This data set contains DNA sequences of the ammonia monooxyenase genes (amoA) from betaproteobacterial ammonia oxidizers. Sequences were obtained from plots located 5 m from the marsh edge at one oiled and one unoiled site within three regions: sites 3 (oiled) and 4 (unoiled) in Terrebonne Bay, LA (TB) sites 2 (oiled) and 3 (unoiled) in Western Barataria Bay, LA (WB) and sites 1 (oiled) and 2 (unoiled) in Eastern Barataria Bay, LA (EB).

Purpose:

This dataset was developed as part of a research project investigating the effects of the Deepwater Horizon oil spill on salt marsh biogeochemistry and microbial ecology. This particular project was directed to determine: 1) if there was a significant impact on community composition of ammonia-oxidizing bacterial communities; and 2) if there were spatial (regional, within marsh) patterns in their distribution.

Methods:

Clone libraries were generated from samples located 5 m from the marsh edge using primers amoA1F and amoA2RTC. Sequences were obtained using T3 primers and Sanger sequencing.

Instruments:

Samples were analyzed at the High Throughput Sequencing Solutions at the University of Washington (Seattle, WA).
