Abstract:

This study was done to evaluate changes in indigenous microbial communities from coastal marsh sediments after the Gulf of Mexico oil spill in 2010. This study was conducted from eleven coastal marshes in southern Louisiana. Samples were acquired from May 2010 through August 2013, focusing on sediments approximately 1 m offshore from the edge of the marsh. Marshes were oiled from late June-August 2010, and some marshes were reoiled in 2011 from tropical storms and hurricanes. Some marshes close to the Mississippi River were impacted by freshwater diversions in 2010. Bacterial communities were assessed from sediments at 0-1 cm and 1-2 cm depth intervals. The results are amplicon pyrosequence libraries of single-direction reads generated using multiplexed, bar-coded primers targeting the V1-V3 region of the bacterial of 16S rRNA gene.

Purpose:

This study was done to evaluate changes in indigenous microbial communities from coastal marsh sediments after the Gulf of Mexico oil spill in 2010.

Methods:

Total nucleic acids were extracted in triplicate. Five grams of sediment for each sample were subjected to methods modified from Somerville et al. (1989; Appl Environ Microbiol, 55(3): 548-554) and Zhou et al. (1996, Appl Environ Microbiol, 62(2): 316-322) that incorporated sucrose lysis buffer with lysozyme (1 mg/ml), a solution of 5X proteinase K/CTAB/SDS, and overnight incubation at 550 C while shaking at 100 rpm. After protein precipitation, nucleic acids were precipitated in isopropanol overnight at -20°C prior to washing with ethanol and purification. Aliquots of triplicate DNA extractions for each of the samples were homogenized and purified for pyrosequencing of the V1–V3 hypervariable region of the bacterial 16S rRNA genes using GS FLX Titanium technology (Roche 454 Life Sciences, Branford, CT, USA).
