Abstract:

Six commercial vessels and two FIO vessels collected fish eggs on the west Florida Shelf on 2017-07-17. At each station, two types of plankton net tows were conducted to collect eggs for proportional species-composition determination via DNA barcoding, and for estimating the number of eggs under the water surface. DNA barcoding was based on the region of the cytochrome c oxidase 1 gene (COI), which was amplified and sequenced using established methods. Individual eggs were identified using DNA barcoding. Sequences were submitted to NCBI GenBank (https://www.ncbi.nlm.nih.gov/genbank/) under accession numbers MK976037-MK97664.

Purpose:

DNA barcoding of fish eggs in the Gulf of Mexico to inform fisheries managers of spawning locations of economically and ecologically important species.

Sequence ID (species, fish egg, and station number); note (Fish eggs identified to mitochondrial barcoding gene- COI, Closest taxa match: % ID, Number of fish eggs processed); species ID (closest species identification); number of fish (number of fish eggs matched to this ID); species probability of placement (probability the species is a match); notes; sequence (DNA sequence); tissue type (fish egg); collection date (date egg collected by plankton tow); geo loc name (station number); lat long (latitude and longitude); isolation source (general collection location); fwd primer name; and rev primer name.

Methods:

At each station, two types of plankton net tows were conducted: (1) a single, horizontal tow that was intended to collect eggs for proportional species-composition determination via DNA barcoding, and (2) three replicate vertical tows that were intended for estimating the number of eggs under 1.0 m-2 of water surface. The horizontal tow consisted of a single, 15-min tow at idle speed using a 335 μm mesh, 1-meter mouth, conical, flowmeter-equipped plankton net. Samples were preserved in 70% isopropanol in ambient seawater. Upon return to the lab, fish eggs within the plankton-net samples were counted and removed using stereomicroscopes. DNA barcoding was based on the region of the cytochrome c oxidase 1 gene (COI), which was amplified and sequenced using established methods. Up to 150 drifting eggs were barcoded from each station. If the total number of eggs in the sample exceeded the number barcoded, the remainder were prorated according to species proportions in the barcoded sample. Individual eggs were identified using DNA barcoding. Sequences were submitted to NCBI GenBank (https://www.ncbi.nlm.nih.gov/genbank/) under accession numbers MK976037-MK97664.
